My computer shows an "E" drive which is not there. I cannot do anything with the icon, like delete it, add more files or remove files.The contents look like a backup CD I burned some time ago and the size is around 612MB which matches a CD. How did this happen and what do I do? I need the drive letter!!!

Perhaps you have Daemon Tools installed on the PC and it's a leftover virtual drive.
See if Daemon Tools is installed and uninstall it.

Or:
Assuming you are running XP:
Start > Control Panel > Administrative Tools > Computer Management > Disk Management
If the drive appears in the window, do a right click on it and choose "Drive letter" > "Change Letter"
Instead of changing the letter, select "Remove" (make sure it's the correct phantom) and see if you can get rid of it.
Reboot to verify it's gone.

The settings for Win 7 are different.
