DAR's and Aspect Ratio's

Can someone please SIMPLY explain what I need to know to get the DAR and/or Aspect Ratio's right when authoring , to get a decent size pic on a 16:9 tv , preferably fullscreen ?
If I have an mpeg2 (see other post) with DAR 4:3 is it too late to change this , when should the DAR be set if transcoding from say an AVI ?
