Do I have to put the movie on my hard drive or can I just open disc and strat DVD Shrink from the disc? I have AnyDVD. Which one is better, on the hard drive first or not?

I too use AnyDVD and DVDShrink and DVDdecrypter or imgburn to burn.
I have done it both ways. You certainly do not need to rip the DVD to the HD first prior to using shrink on the files. I have done plenty of backups using AnyDVD, DVDShrink and then the burn program. Works great.
What I found, with my system is that it tended to take longer for Shrink to do its thing going direct to the reader to gather the DVD files than if I ripped to the HD first. Consequently, I have went back to ripping to the HD first, then shrinking, and burning lastly.
I would have to agree with ruger. I've gone both routes and typically rip to HDD first with DVD Decrypter, because DVD Shrink usually takes longer to rip from the disc than the hard drive...but the time saved is usually not substantial, maybe 4-5 minutes depending on the movie.
Anywhoo, neither is the wrong way, its your choice. So do what feels best to you.
