Can I copy straight from retail disk to backup?

A bit new to this making backup's thing.....

Having just purchased a new dvd, can i just make a straight copy onto dvd -r?
I have a program called DVD43 installed which claims to overcome the protection issues, but not sure what program to actually use to make the backup?
