unless you have a mod chip that can play DVD9 backups (DMS4 Pro)
or a modchip that can play regular original DVD9 games and you patch the back up with Toxic DL Patcher then you can't
because you would have to burn that to a DVD9 (dual layer) disc
the only other way is to find a guide somewhere that will show you how to rip that game from a DVD9 down to a normal DVD5 size, which requires you to remove a lot of extra's and cutscenes and such

The Iso looks like it may have False LBA so you maybe able to shrink it to fit a DVD5
Try processing the Iso through Experimental Plugin Ext Reb Tool dvd iso shrinker and see what happens

here is a link to Toxic DL Patcher in case you want to
just burn the whole game to a dual layer disc and not shrink it.
http://www.teamtoxic.com.cn/
it's at the very bottom of the page.

cut it out with all the help help help :(
converting it to iso was wrong, the game is dual layer, what you have there is one layer
what chip have you got?
and while i'm here, just to clear a few things up about MGS2 substance
1)if you havent got a modchip that boots DVD-9 backup, there is no other way to back it up without splitting it onto 2 dvd disk's
2)there is no false lba,so shrinking it that way is not a option
3)there are numerous DVD-9 checks which need some manual hex-editing,and only the NTSC version, has a guide written showing you which strings to edit (you cannnot use console media patcher)
4)if you have the PAL version, search around, get in contact with Gnie, and if you are nice,he may send you the patched files that you need

it dont matter whether you ps2 is NTSC,if you have a PAL version of the game, you wont beable to rip it.
all ps2's boot DVD-9,its just the backups that most cant boot.
if you are confident that you can boot DVD-9 backups then just use a DVD-9 disk like suggested by orthogonl

well, you should be ok then if the game is NTSC
@tmfloria,yes you are right,;) there is a "kinda" rip-kit out there, its sorta useful if you dont know how to hex edit the slus ioprp.img file
as those files come with it.
but there is no full rip-kit,you still need to do a hell a lot of the work yourself.
good luck!
