Is it must to have lumines UMD to downgrade my psp or i can use lumines ISO file. i hav psp ver 3.30 ??

You need the UMD.

No, you can just go against everything the Readme clearly explains to you. You can also run an ISO on 3.30/3.50 without downgrading making it completely pointless to downgrade anyway.
