If My PAL Wii is fully up to date 2.2E (I believe) and online, if I put in a Super Paper Mario NTSC-U disk will it brick my system?

Also Should I use RegionFrii First?

Why don't you use the Wii Brick Blocker to patch it just in case? Search for the program on this site.
