Did you try to press the subtitles button one more time to show "on"?
If you play the Dvd that you just made in your computer, can you turn the subtitles on? If you still have the folder that Convertx2DVD made, just play the VIDEO_TS.IFO file from the Video_TS folder(you can use VLC )

Can you see the subtitles when you play the MKV or the DVD?

Were you ever able to play a DVD off the the USB, with menus/subtitles? I am sure it works if you use an actual discs, but might not work from the USB.
Anyhow, your player specs say it will play MKV and Divx. Just copy the MKV file to your USB and should probably work.

The MKV will not work? It says it should, it's supported.
If you want to play off USB, try and convert the MKV to an AVI(Divx) instead of DVD.

Ah, it would be so much easier than convert to DVD.4 hours??? What computer do you have? It definitely should not take more than converting to DVD. However, I would still look into figuring out why MKV don't play. Get the manual out, maybe find some specifications, or if you feel brave, call tech support.
Try this to convert to Divx:
http://www.any-video-converter.com/download-avc-free.php
Choose AVI for container, choose Xvid for codec. Since it can play HD Divx, you can choose a higer frame size, like 1080x720, or even maxim HD, and bitrate, 1024 or so.It can hardcode the subs, so you don't have to worry about them.
Here's a forum where you might be able to ask and see why you cannot play MKV(I am curious as to why it doesn't work).

I just glimpsed at some news, and it might be possible that your player supports MKV, however, needs to be encoded using Divx. Most MKVs available on the net are encoded using H.264, that's why your player might not be able to see it. It makes sense, since H.264 capability is not mentioned.
So instead of AVI, just choose MKV.

No, just forget that, my bad. I missed some important detail, the "Plus HD", that your player does not support.
Just make an AVI using xvid like I mentioned.

After you open the video, scroll down, below the video frame/bitrate...settings and choose the subtitle.
The subtitle will become part of the video, you will not be able to turn them on/off. If you want to be able to switch them on/off, then don't select them, is another process.
The quality is always degraded to some level every time you do a conversion, even when converting to DVD.

A quad core should do it in no time.It takes 30 min or less on a core2 for me. Just make sure you set the proper number of cores(set to "4" the "Number of threads used for encoding":
