Little help please, this may be the software, it may be the emulator or it may be the operator

I bought the collectors edition PC game and have been using a backed up copy to keep my real one in pristine condition (and cause my DVD drive is tempremental as hell)
I run the game via Daemon tools and I had around 80 hours played when it crashed badly.
Now when I go to run it again it tells me that it can't find the game exe and that I need to re-install. I did that but it keeps telling me it can't find the file. I have re-installed the software and Daemon tools but still no joy. Any ideas?
