I tried backing this game up on a DVD using this guide on DVD encryptor:

Well, my PS2 doesn't want to play it and I have no idea why because the burning went smoothly. When I downloaded it it was a .rar file and then I extracted it and tried to burn the MDS to a CD which was succesfull but my PS2 refuses to play it. It flashes green lines on the screen and then I get to the memory card screen... My Ps2 is chipped and it plays other back-ups great (not when I try it though). What is the problem? I used a Verbatim 16x DVD+R cd and burned with 8x speed. Could the whole file be corrupt and if that is the case, how do I find out?

I meant that it doesn't work when I try to make a back-up disc, but when others do it they are succesfull...

Dark Cloud 2 is a DVD-ROM game. Unless you downloaded a CD rip of it, you should burn it to a DVD. 16x media is iffy with PS2 backups, best to stick with quality 8x media like Taiyo Yuden and Verbatim. How big is the file you're trying to burn?

I used Verbatim DVD+R, however they are 16x...
The file is about 4.3 gb.

I tried backing it up again using this guide to further enhance my burner, and now it works!!

I also lowered the speed down to 6x when I tried burning it again and now I have a fully functional back-up of Dark Cloud 2 on my shelf :D

Glad you got it working. That guide you were using is for PSX/PS2 CD-ROM games, use the guides in my sig for DVD-ROM games in the future.
