To make an ISO you'll need the files from the CD and then IMGBurn to make an ISO of it and then burn it.
However, bear in mind you won't be able to play the backed up game on your PS2 without some sort of modding.

hi. i have files that i copied from a game cd that i dont have anymore, it broke. so pl tell me how can i make an image of those copied files so that i cud play it on virtual drive.

If you have copied those files directly from the CD, then all you need to do is use the guide Ripper linked to build an .iso from them.
If you already have the game installed, then playing it off a virtual drive should not be a problem, however some games don't like this and require a bit of a "helping hand" to get working.

i hav the game installed but it gives error "insert cd rom" that i don hav no longer. It is gangsters 2 v1.0.4 english[old but i like it].

Is your PS2 modded? What speed did you burn the disc at? What media are you using?
Forget the above reply and see the edit in this post; I was replying to this and a thread where member was trying to back-up a PC game at the same time, got confused between the two and posts got mixed up, sorry.
