I've recently read the guide on backing up your game and adding the DNA's to enable online play. I was sure I read that it was possible to use this method on your hard drive, but am unable to find it.

I thought that the server checked your setup and when it detected am unofficial hard drive it would not allow access. Could anyone supply any info on this? it would be greatly appreciated!!

no, you cant play online off the hd with dnas enabled games right now. I did read about a group working on it, but only a few games worked and i haven't heard any more about it.
