The rear cover of the disks case will display if the movie is in a PAL or NTSC format. If you just have movie files and want to know the format, then the apps suggested will do.
Also, you can Load it into GSpot (if its an AVI, mpg or VOB) and if its frame rate is 25.0, its PAL and 23.976 or 29.97, its NTSC

PAL and NTSC go by region. If you know the region, just check to see if the region uses the NTSC or PAL standard. For instance the US uses NTSC and is part of Region 1. The UK is part of Region 2 and PAL is used there. Which means Region 1 = NTSC and Region 2 = PAL. You can google for world maps with Region codes and NTSC/Pal usage. It doesn't take anything special to check the format, just check according to region. If not from your region, the PC will indicate a problem by opening a Confirm Hardware window. Don't manually change any settings for the drive as far as regions go, leave it set to the home region because you have a small limited number of changes before it becomes permanent. You can use DVD Region + CSS Free or AnyDVD to bypass region protection and it will give the region code as well. As mentioned factory DVDs are usually labeled as far as region and NTSC or PAL format go. If a home DVD, you don't need to worry about region codes, but if you know the region origin it's easy to determine format.
