What DVD are you trying to rip and burn? If it's a movie you're trying to back up it probably still needs to be decrypted, which shrink won't do. You need a decryption software...like DVDFab HD Decrypter.
Here's a guide to help you get started

it is but its a very poor program . I would uninstall it and use either dvdfab decrypter(freeware) or use anydvd

What version of Nero are you using?
Not reading media can be a drive-media issue. What is the drive, is the firmware up to date, and what media are you using? If not sure of the manufacturer, it is listed in Nero burn logs or you can use the easy freebie tool DVD Identifier to find out.

Yeah it sounds like something to do with the drive. Whether it's the media not being recognized by the drive or something wrong with the drive itself, we need some more info.

As you can see in the photo below, I have a PLEXTOR DVDR PX-716AL selected to burn and an extra drive that is an HP DVD Writer 740b. You should have one drive listed. When you open the Shrink Backup window, what is showing below Select backup target: ? Would you post that for us?
